Abstract

A cell culture which produces a desired polypeptide is prepared by genetic modification of microorganisms. The process comprises the following steps: a) splitting up molecules of a DNA receptor material obtained from a plasmid which is suitable for the infection of cells of the said microorganism, by treatment of the plasmid DNA with an endonuclease. b) Splitting up another genetic material which contains DNA donors and codes for the synthesis of a desired polypeptide, using a similar endonuclease, resulting in DNA fragments whose ends are compatible with those obtained according to a), c) mixing the DNA fragments obtained from the various sources, resulting in a random attachment of the DNA fragments under the influence of a DNA ligase, which results in hybrid DNA molecules which contain the combined fragments from both DNA sources, d) infecting the microorganism cells to be modified by contacting with the hybrid DNA molecules, e) allowing the infected microorganism to metabolise in the presence of a substrate which is suitable for conversion into the desired polypeptide and f) screening for production of the desired polypeptide and preparation of a cell culture which produces the polypeptide. Preferred applications are the production of mammalian polypeptides and proteins, the production of bacteria by "foreign" enzymes and secondary metabolites, e.g. glucose isomerase, and the production of compounds such as citric acid and cephalosporin by modified tissue.
