Abstract
A method for obtaining a modified microorganism for containing a nucleotide sequence with the structure of a gene of a higher organism, transcribed from that gene, which method comprises: a) isolating cells of a higher organism containing mrna that encodes the desired gene; b) extract the mrna from the cells in the presence of a rnasa inhibitor composition, thereby preventing mrna degradation by rnasa. C) separating the mrna essentially free of protein, dna and other rnas, d) synthesizing a cdna or double helix, where the chain contains a nucleotide sequence complementary to that of the mrna, thereby producing cdna with a nucleotide sequence encoding the desired gene, e) provide a dna transfer vector with reactive ends capable of being linked together or of being joined with double-stranded cdna, f) attaching the dna transfer vector to the double-stranded cdna containing a sequence of nucleotides encoding the desired gene, and g) combining a microorganism with the vector of dna transfer bound to the cdna with a nucleotide sequence encoding the desired gene, thereby producing a microorganism with a nucleotide sequence encoding the gene wanted.
