Solute carrier family 35 (SLC35) members transport nucleotide sugars and adenosine 3'-phospho 5'-phosphosulfate from the cytosol to the endoplasmic reticulum and the Golgi apparatus. DE NOVO GDP-FUCOSE BIOSYNTHESIS -
In the 'de novo' GDP-L-fucose biosynthetic pathway, GDP-L-fucose is produced from GDP-D-mannose via a 2-step process in the cytosol and then imported in the Golgi apparatus where fucosyltransferases use it as a fucose donor for the N- and O-fucosylation of proteins and lipids.This is the only pathway for GDP-L-fucose biosynthesis in D. melanogaster as the enzymes involved in the salvage pathway - which uses L-fucose as a substrate - are not present in the Dmel genome. Involved in GDP-fucose import from the cytoplasm into the Golgi lumen (PubMed:15530860, PubMed:22745127). Plays a major role in the fucosylation of N-glycans (PubMed:15530860, PubMed:22745127). Functions redundantly with Efr in the O-fucosylation of Notch, positively regulating Notch signaling (PubMed:19948734).

Phenotypic Description (Red Book; Lindsley and Zimm 1992)
nac: neuronally-altered-carbohydrate (F. Katz)
Mutants show alteration or loss of a normally neuron-specific glycoconjugate; staining by anti-HRP antibodies in imaginal and adult neural tissue is eliminated. At 25 the mutant flies are viable and fertile; nac/nac females, however are sterile at 18. Under heat stress (37 for five minutes) they show abnormal jittery behavior. Developmental abnormalities, including defects in the assembly of the ommatidia and in the formation of the wing, appear at 18 in the homozygous offspring of heterozygous parents. Maternal effect embryos show loss of the anti-HRP glycan determinant and are lethal.

        Gfr may transport GDP-fucose from the cytosol into the golgi.

        N signaling deteriorates in Gfr mutants.

        The phenotypes of Fas1, Fas2, Fas3 and nac mutants were analysed in the developing wing: the axon tracts in the CNS for the most part remain unaltered, and none of the phenotypes are 100% penetrant, indicating a fine-tuning role for these molecules in both the PNS and CNS.

        A mutation in nac affects anti-horse radish peroxidase (anti-HRP) epitope expression in imaginal nervous tissue causing behavioural defects.
