Lissencephaly-1 (Lis-1) encodes a regulator of the cytoplasmic dynein-1 motor complex, binding to Dynein heavy chain and regulating dynein force production, microtubule affinity and velocity. It is involved in the transport of many of dynein's cargoes, including mRNPs, mitochondria and nuclei. The Lis-1 product is also a non-catalytic subunit of platelet-activating factor acetylhydrolase. POSITIVE REGULATORS OF BMP SIGNALING PATHWAY -
Positive regulators of Bone Morphogenetic Protein (BMP) signaling up-regulate the pathway, ultimately resulting in the increased nuclear activity of the Mad/Med transcription factor complex.
Protein Function (UniProtKB)
Positively regulates the activity of the minus-end directed microtubule motor protein dynein. May enhance dynein-mediated microtubule sliding by targeting dynein to the microtubule plus end. Required for several dynein- and microtubule-dependent processes such as nuclear migration during cell division, mitotic spindle formation and the removal of mitotic checkpoint proteins from kinetochores at the metaphase to anaphase transition. Required for several aspects of neurogenesis including neuroblast proliferation, neuronal cell differentiation, dendritic growth, branching and maturation and axonal transport. Required for synchronized cell divisions in the germline, fusome integrity and oocyte differentiation. Acts together with BicD, Egl, dynein and microtubules to determine oocyte identity during oogenesis. Also required for nurse cell to oocyte transport during oocyte growth and for the positioning of the nucleus in the oocyte.

        Lis-1 acts as a negative regulator of mitochondrial transport along the axons in aging adult neurons.

        Lis-1 protein is recruited to RNA localisation signals and is required for net minus end-directed transport of apically localised RNAs. Lis-1 promotes the recruitment of dynein and dynactin to RNA localisation complexes. In addition, Lis-1 protein levels appear to regulate the overall association of dynein with dynactin.

        Lis-1 controls germline stem cell self-renewal by maintaining BMP signaling in germline stem cells and shg accumulation in the stem cell-niche interface.
