UASt regulatory sequences drive expression of the sl open reading frame (amplified from the RE62235 cDNA clone). The ORF has been mutated to carry a D1184G amino acid substitution. This change is equivalent to a D1165G change in the orthologous human PLCG1 gene, a putative pathogenic variant. The coding sequence is tagged at the C-terminal end with three copies of Tag:HA. The ORF is flanked by a pair of incompatible FRT sites (FRT5 and FRT2), which allows for future in vivo exchange of either the promoter or tag sequence.
