Expressing shotJF02971 under the control of Scer\GAL4SPARC-MI00329-GAL4 results in third instar larval adipocytes presenting plasma membrane invaginations where aberrant extracellular matrix material accumulates (Vkg.GFP), despite of no obvious secretion defects (secr.GFP).

Expressing shotJF02971 under the control of Scer\GAL4Cg.PA results in third instar larval adipocytes presenting enlarged, deformed and highly eccentric nuclei, plasma membrane invaginations where aberrant extracellular matrix material accumulates (LanA.GFP, LanB1.GFP or Ndg.GFP). In agreement, Golgi units lose their homogeneous distribution throughout the cytoplasm and seem to gravitate toward a single focus near the nucleus. These adipocytes also exhibit a large juxtanuclear focus of actively growing microtubules, in contrast to a normal perinuclear distribution in controls. This ectopic microtubule focus concentrates actin filaments. The long tubular extensions are reminiscent of a mitotic cytokinesis furrow, although no mitosis (anti-Phospho-H3) can be seen. There is no effect on adipocyte ploidy.

Conditional expression of shotJF02971 from the L2 under the control of Scer\GAL4Cg.PA and Gal80[ts] also leads to aberrant matrix accumulations (vkg.GFP) and concentration of microtubules in a large juxtanuclear focus: 36h induction leads to an incipient microtubule focus abutting the perinuclear network; 48h or 60h induction causes more dramatic microtubule single point concentrations, similar to continuous expression.

Expressing shotJF02971 under the control of Scer\GAL4rn-GAL4-5, Scer\GAL4pnr-MD237 and Scer\GAL4byn.PU caused both aberrant microtubule foci formation and mispositioned nuclei in the salivary gland, larval epidermis, and rectal papilla, respectively. The epidermis and salivary gland juxtanuclear foci exhibit non-centrosomal minus-ends (Patronin.GFP).

Expressing shotJF02971 under the control of Scer\GAL4NP0001 leads to the concentration of microtubule minus-ens in a juxtanuclear focus in third instar larval midgut enteroblasts.
