Pp1-87B87Bg-3 Pp1α-96A2 double heterozygous embryos exhibit damped (and spatially more confined) oscillations in the Cdk1/PP1 activity ratio, reduced cortical contractions and cytoplasmic flows, non-uniform positioning, tissue-wide asynchronies, and anaphase delays when compared to controls.
Eye morphogenesis is disrupted and eye size is reduced in animals containing eyes that are almost completely homozygous for Pp1-87B87Bg-3. In addition, mutant photoreceptor cells, cone cells and undifferentiated cells are mislocalised in the optic stalk of these animals.
Pp1-87B1/Pp1-87B87Bg-3 animals die during pupariation, and until pupariation, do not lag behind in development compared to their control siblings.
Pp1-87B87Bg-3/Pp1-87B87Bg-6 larvae show axon clumping at the lamina and laminal breaks with ~50% of R2-R5 axons being mistargeted to the medulla.
Squash preparations of Pp1-87B87Bg-3/Df(3R)kar-H10 third larval instar brains contain fewer cells per microscope field than normal and a significantly higher proportion of cells in mitosis than in wild type. Overcondensed chromosomes are seen.
Weak suppressor levels of position effect vareigation and a high frequency of aberrant mitosis in neuroblasts or ganglion mother cells. Metaphase chromosomes and anaphase chromatids are over condensed.
Larvae die either just before or just after pupariation. Third instar larval imaginal discs are underdeveloped, indicating failure of diploid cell proliferation. Ganglia are smaller than those of wild-type larvae, with significantly fewer cells per microscope field in squash preparations, suggesting cell division is impaired in the ganglia. The proportion of cells in mitosis in the ganglia is significantly higher than in wild type. This is due to a large number of mitotic cells showing abnormalities, such as overcondensed or extra chromosomes. These cells are probably delayed in progress through mitosis. Spindle organisation is also defective, showing unusually little space between almost parallel bundles of spindle microtubules at metaphase.
